Current estimates have found that up to 36% of all cell lines in use are either misidentified or cross-contaminated (3). Authentication testing is meant to be used as a routine quality control check, to verify the identity of a human cell line being used for various experiments and for publication and/or grant funding. The validity of the experimental data often depends on the identity of the human cell line(s) in use.
Currently the gold standard for human cell line authentication testing is a technique called short tandem repeat (STR) profiling. STRs are repeated segments of DNA that are typically 2-6 base pairs in length and are scattered throughout the human genome. Our STR profiling test utilizes PowerPlex16HS and includes 15 autosomal sites, plus amelogenin (gender marker), as well as a mouse marker for detection of mouse DNA contamination (especially helpful for researchers creating human tumor xenograft models utilizing mice).
